While the neurological and vascular aspects of Hand-Arm Vibration Syndrome (HAVS) has been generally accepted as a medical condition, the medical criteria and the clinical findings used to establish the diagnosis has been more difficult to bring to consensus. The criteria was first quantified by the Taylor-Palmear scale.1 This criteria was subsequently modified in 1986 at the 1st Stockholm Workshop2,3 to included more acceptance for the neurological effects that characterized the predominate findings in some workers. The relationship between hand-arm vibration and Carpal Tunnel Syndrome was defined in NIOSH 97-1414. While the aforementioned documents have defined the clinical entities associated with hand-arm vibration exposure, agreement on the clinical findings and test to confirm the diagnosis has be been more difficult to bring to consensus. Clinicians assessing HAVS has relied on a number of varied neurological and vascular tests. The neurological testing has focused on assessing damage to the sensory capability of the fingers for the neurological component including tests to measuring ability to sense vibration, cold or other end point finger sensor functions. However, the vascular testing has been traditionally focused on the ability to either measure vascular function or to reproduce the vascular blanching that occurs in HAVS with cold water provocation. Recent assessment of this testing in the United Kingdom Coal Miner’s study has questioned the value of this testing especially in reviews by McGeoch.5 In an attempt to provide some type of definitive testing to substantiate vascular damage from hand-arm vibration exposure, angiography is an alternative or adjunct to cold water provocation testing. The standards that have been established to predict the level, type and incidence of HAVS have been based on clinical studies and reports that have essentially been all male populations. However, the recent entry of women into more vibration intensive jobs has brought about the exposure of some women to high levels of vibration previously only previously experienced by men. However, there have been only few studies that look at HAVS in women6. Although exposed the same vibration levels, it has not been clear that the latency and type of pathology of HAVS in women will be the same as for men. The purpose of this study is to look at recent case studies of men and women exposed to jobs with high levels of hand-arm vibration with extensive clinical testing for both the neurological and vascular components of HAVS as well as other associated upper extremity conditions such as Carpal Tunnel Syndrome.

The overall disabling work injury experience, excluding office- workers, at nometal mines and mills during the period 1961-63, as reported to the Bureau of Mines by operators, was 77 fatal and 7,201 nonfatal injuries during an aggregate worktime of 335,029,685 man- hours. These injuries, fatal and nonfatal combined, occurred at a frequency rate of 21.72 per million man-hours, comprising 0.23 for the fatal and 21.49 for the nonfatal injuries. The combined severity rate of the disabilities was 2,312 days lost or charged per million man-hours, consisting of 1,379 for the fatal and 933 of the nonfatal injuries, The average severity of all disabilities during 1961-63, was 106 days per injury, The number of days lost or charged, excluding officeworkers, at nonmetal mines and mills during 1961-63, was 462,000 days charged to fatal injuries and 312,461 days lost from nonfatal injuries. The outstanding causes for fatal injuries were explosions (18 fatalities occurred in a major mine-explosion disaster in a potash mine in Utah in 1963) and falls of rock or ore from back or side, while most nonfatal injuries were caused by handling materials, slips or falls of persons, haulage, and machinery. The parts of the body most frequently injured during the period 1961-63 were the trunk, hand and finger, and foot and toe, An average of 52,810 men were at work daily, There were 20 nonfatal disabling injuries reported for office- workers at nonmetal mines and mills during the period 1961-63 during an aggregate worktime of 23,088,496 man-hours, These injuries occur- red at a frequency rate of 0.87 per million man-hours, There were no fatal injuries reported.

The Bureau of Mines studied waste disposal methods a1 a coal mine in Kentucky and another in Alabama to develop cost estimates, to provide better knowledge of current practices, and to guide consideration of alternate disposal methods. Costs of the existing waste disposal systems at each mine were estimated using present-day costs of equipment and labor. Costs of waste disposal systems using alternate equipment were also estimated. Although total disposal costs were essentially the same for both operations, reclamation costs will differ because of different reclamation practices.

"INTRODOCTIONIn an earlier communication 1 from our laboratory it was reported that mechanical crushing of coal and quartz under normal air atmosphere generates of free radicals on the particle surfaces, and that these radicals decay with time, hence pointing to a higher toxicity of fresh dusts in relationship to pneumoconiosis and silicosis. More recently Fubini et al.2 have also reported the detection by electron spin resonance (ESR) of the formation of SiO. and Si-type of radicals from quartz particles crushed under atmospheric conditions. In agreement with earlier ESR studies on single crystals of quartz crushed under high vacuum ( -10-10 torr>3 and subsequent exposure to air,3 and to other gases,4 these radicals were identified2 as being formed by the homolytic cleavage of the Si-0-Si bonds and the reactions of the Si• and SiO- radical with atmosphere. Fubini et al.2 also suggested that these radicals might be involved in the mechanism of the fibrotic action by silica, either by transforming the particle surface into a selective oxidating agent or as an initiator of a sequence of reactions leading to fibrosis. Earlier Gabor and Anca5 bad reported that lipid ·peroxidation caused by free radicals on the silica surface might be involved in the red blood cell membrane damage. Thus far, however, no parallel cytotoxicity, fibrogenicity, and free radica1 studies on a given quartz dust sample have been reported, except for some earlier work from our laboratory.1·6·7 We now present more recent results obtained from parallel cytotoxicity, fibrogenicity, and free radica1 measurements on a freshly made quartz dust. The dust's free radica1 content was measured using ESR spectroscopy while its cytotoxicity potential was estimated via hemolysis. Hemolysis was employed as the toxicity test because it is a widely used method for estimating the potential of a dust for disrupting the cell membrane. 8 The fibrotic potential was followed by measuring the dust-induced lipid peroxidation, using linoleic acid as a model lipid. As discussed below, the results obtained suggest new clues to the mechanism of the quartzrelated cytotoxicity and fibrogenicity. MATERIALS AND METHODSReagentsCrystalline quartz particles with a size range of 0.2-2.5 mm were obtained from the Generic Respirable Dust Technology Center, Pennsylvania State University, University Park, Pennsylvania. These particles were crushed in air to obtain quartz dust samples with particle sizes smaller than 20 microns. We chose to work with a dust with mixed particle sizes, rather than a specific range, as an effort to simulate the mining atmosphere. An agate mortar-pestle arrangement was used for the crushing and grinding because of the close similarity of the structure of agate to that of quartz. Diethylenetriaminepentaacetic acid (DETAPAC) were purchased from Sigma. All other chemica1s were purchased from Fisher or Aldrich.Hemolysls ExperimentsHemolytic activity of silica was measured, following an established procedure,9 as the amount of hemoglobin released from a 2 % suspension of sheep erythrocytes after incubation with 10 mg of silica dust for one hour at 37 °C. The hemoglobin release was estimated via the absorbance at 540 nm using a Gioford spectrophotometer. The procedure was calibrated by substituting the silica dust by a phosphate buffer solution as a negative control (background) and O.S % Triton-X-100 as a positive control (100% hemolysis). As mechanical shock and vibration environments evolve, it is important to understand their potential effect on human operators. Human beings are sophisticated mechanisms comprised not only of passive components with mass, damping, and stiffness characteristics, but also of components that can actively affect apparent mass, stiffness, and damping. Because the lumbar spine can exhibit local, short-column buckling, stability of the human trunk depends on the responsiveness of the neuromuscular control system.1-2 We have been evaluating the ability of patients with back pain to respond to a series of sudden loads. We believe the results have implications for isolation design and standards development. Methods 153 patients, aged 21 to 55, presenting with back pain agreed to enroll in a research study that randomly assigned them to one of three treatment arms: high velocity low amplitude spinal manipulation, low velocity variable amplitude spinal manipulation, or wait for 2 weeks and then be randomized to one of the above groups. Response to sudden load testing was one of a battery of baseline evaluations performed upon entry into the study and prior to treatment. EMG electrodes were attached to the skin over the paraspinal muscles of the standing participant bilaterally 3 cm from midline at the L3 level. While standing upright on a force plate (Bertec), participants were fitted with a strap around their back and hooked to a load cell in front of their chest. An accelerometer was rigidly attached to the load cell. Impact was applied to the chest using a cord attached to a falling weight. The weight’s fall distance was varied between 9 and 13 inches to account for the size of the subject. The subject was blindfolded and wore headphones playing white noise to prevent cueing of when the weight was dropped to apply the load. Hence, although the participant knew a load was about to be applied, he or she did not know the instant it would occur. Just before the weight was dropped, a 4 second data collection process was started for the two EMG electrodes, load cell, accelerometer, and force plate. The load drop was repeated 6 times, at irregular intervals, over a period of 2 minutes. The raw data thus collected was reduced to obtain several values: 1) length of time from the pull on the harness to the beginning of the response of the left and right paraspinal muscles (LES, RES), 2) time and magnitude of the maximum response, 3) force and acceleration experienced at the chest, and 4) the time and magnitude of the center of pressure location (COP). A general linear model was used to evaluate the results. Results For the EMG data, of the 1,824 observations made, 90% of them indicated a response. Prior to the sudden load, resting muscle activity was different between left and right sides (p=0.0001) and between males and females (p=0.0001). Female subjects began to respond to the sudden load within 92 to 110 ms and males from 101 to 109 ms. Females exhibited more variation in starting

Analysis of the helium content of a natural gas presents more than the usual difficulties encountered in applying the thermal-conductivity method because of the relatively small amounts of this inert as present and the wide variation in the other constituents. However, the numerous advantages of this method and its adaptability to Continuously recording and control instruments make it worthy of consideration. A specific application of the method described yields quantitative results only when used with gas mixtures of known qualitative compositions which Vary within predetermined limits. Periodic analyses for the helium content of a producing gas well may be conveniently made by an absorption method, and a continuums analytical method is not necessary for this purpose. Natural gas containing 1.75 to 1.90 percent of helium produced from Government-owned wells is processed in the Amarillo (Texas) Helium Plant of the Bureau of lanes. The helium extracted in the plant is supplied to the Army and Navy, and the processed natural gas is sold to a commercial gas company. As complete recovery of the helium in the raw material is not economically practicable, the processed gas contains 0.1 to 0.3 percent of helium, depending upon conditions of operation. Production is checked against the helium removed from the gas, which before the installation of the thermal-conductivity apparatus was calculated from the analysis of a cumulative sample of the processed gas. As conditions of operation affect the residual helium content to a large extent, a composite sample was an uncertain index of the efficiency of the process.

"According to accident statistics, issued from time to time by the Department of the Interior, Bureau of Mines covering fatalities in coal mines, between the years 1870 and 1921 there were 4,897 deaths caused by explosives. The legislatures and safety commissions of the various mining States have Elected and formulated many excellent regulations in regard to the handling and use of explosives in an endeavor to keep this class of accidents as low as possible. An analysis or comparison of the explosive, regulations issued by over thirty States would make a very long paper, however, by subdividing according to phases and issuing several papers it is believed that the material will be more readily available. The first phase was treated in Serial No. 2405 of the Bureau of Mines (""State and Federal Electrical Shot-Firing Regulations,"" issued in October 1922) which gave a compilation of such regulations as apply strictly to electric shot-firing. The second phase under consideration is ""Who may set off blasts in coal mines.""In some States, the person who does the firing must be an ""authorized shotfirer,"" while in other States he must have proved his ability to do this work, and again in other States he is ""any person."" The present paper covers rules and regulations relating to this phase of shotfiring, and those States having similar regulations have been grouped as follows."

"INTRODUCTION With the advent of war in 1941, military operations in Alaska were enlarged on such a scale that the demand for coal in the Territory more than doubled. Coal was needed urgently for heating forts and other military establishments, for use on large construction projects, and for handling the greatly increased volume of traffic on the Alaska Railroad. -The additional tonnage required, estimated at perhaps 250,000 tons per year, probably could have been supplied by the United States, but shipping space between Pacific Coast ports and Alaska was already overburdened with military supplies and construction materials. Moreover, although of only secondary importance in wartime, the cost of coal shipped into the Territory is reported to have been as high as $40 per ton. The alternative to importing coal was, of course, to increase the Production of the Alaska mines.Production of coal in Alaska reached, 350,000 tons in 1944, or more than double the prewar' output. The responsibility for increasing coal production was assumed largely by the Army, A Coal Commission attached to the Alaska Department arranged financial assistance for some mining operations, aided the operators in producing equipment for mechanization, and provided most of the labor necessary to operate the mines. The Bureau of Mines contributed to the program in several ways. First, exploration by the Bureau in the Moose Creek district revealed a substantial tonnage of coal of better grade than any currently mined in the Territory, 4/ and drilling in the Broad Pass district, where a mine to supply coal for the army had been started, revealed that the reserves were far less than had been supposed; this operation was abandoned. Second, coal-preparation practices were studied, and recommendations for improvements made, at two of the operating mines at which the quality of the coal makes preparation a factor of vital importance. Some of the information obtained in these studies forms the basis for the present report."

ackleg rock drills are widely used in the mining industry and are known to generate high levels of hand-arm vibration which contribute to the development of the hand-arm vibration syndrome for exposed miners.1-3 To reduce the vibration levels, a prototype of an antivibration handle was developed as part of a previous study.4 To provide some bench marking for this handle prototype and to follow the evolution of its performance over time, a test bench was developed to characterize the vibration emission values of jackleg drills under controlled operating conditions. As the current ISO 8662 series of standards could not apply directly to this type of tool, there was a need to design and validate a test bench to evaluate the vibration emission values of jackleg drills, while taking into account the conditions specific to the operation of this type of tool. Methods A test bench including an energy absorber, was developed for testing jackleg drills based on the ISO 8662-3 standard 5. The energy absorber was bolted to a 3300 kg concrete block to ensure tool stability. A pictorial view of the device is given in Figure 1. For validation purposes, acceleration measurements at the handle of a conventional jackleg drill were taken simultaneously along the three axes (xh, yh and zh) in an underground rock drilling operation as well as on the test bench. The handle accelerations were measured for three different jackleg angles (13º, 28º and 43º) determined with respect to the floor. Moreover, each measurement was repeated at least three times to asses the data repeatability. [ ] Results As a preliminary validation of the test bench, Figure 2 provides a comparison of the frequency weighted rms acceleration spectrum measured along the zh-axis, for both underground drilling and operation on the test bench (28º jackleg angle in both cases). It is shown that the vibration measured on the handle of a jackleg drill operating on the test bench is representative of that recorded during typical rock drilling operations, despite the fact that some harmonics of the percussion frequencies are generated with a higher amplitude on the test bench. Following the pattern established in the 1989 edition of this study, the concept of data availability must be addressed, because there are perceptible changes from year to year, and the availability of accurately reported, precisely defined statistics and nonstatistical information provides the only sound basis for analytical studies of any type. As the countries of eastern Europe, including the numerous states that formerly comprised the U.S.S.R., undergo the transition from centrally planned economies' to market economies, there has been a perceptible increase in information availability for rather obvious reasons. Any industrial operation seeing economic assistance from government and/or private sources outside the country in which the facility is sited must provide some appraisal of its operations at the time it seeks assistance, as well as some information on previous performance to serve as tools in evaluating its potential under the proposed development scheme for which it seeks financial assistance. This situation, coupled with governmental recognition that concealment of much of the national mineral production, trade, and consumption data for purposes of national security probably never was really necessary and certainly proved unnecessary with the end of the so-called cold war, has led to the publication of increasing amounts of both statistical and nonstatistical information in the countries undergoing economic transition.

Vibration exposure at workplaces is generally multi-axial. The health effects of vibration exposure also likely depend on the vibration direction. Therefore, there is a wide interest in the simulation of multi-axial vibration in laboratory experiments. Advances in technology have led to the development of a new 3-D test system for studying hand-transmitted vibration exposure and health effects. The purposes of this paper are to introduce the system and to present the results of its preliminary evaluations. Test System As shown in Figure 1, the system is basically composed of three vibration generators, a multi-axis vibration control system, instrumented handle, handle fixture, and shaker-fixture linkages (stingers). [ ] Figure 2 shows the array of three vibration generators (MB Dynamics, Energizer BLACK-500 lbs) and their associated support bases and foundation developed by MB Dynamics (Cleveland, USA), which create the 3-axis simultaneous motion. These electrodynamic exciters are powered by power amplifiers which provide current proportional to the analog drive signal from a controller. The controller (JAGUAR Multi-Input/Multi-Output closed-loop vibration controller) was provided by Spectral Dynamics, San Jose, California, USA). NIOSH-designed instrumented handle was equipped on the system.

[The growing importance of the Sundance (Jurassic) formation as a source of oil in the Rocky Mountain area has prompted the making off a series of porosity determinations of core samples collected from the reservoir sands of the Lance Creek oil field, Niobrara County, Wyo. These analyses are preliminary to a study of fluid and energy relations in the Basal Sundance reservoir of that field, and the determinations of porosity will be used in subsequent studies requiring calculations of the volumetric capacity of the reservoir and estimation of the reserves of oil and gas.]

The Bureau of Mines subjected three commercially available brands of transformer fluid to a series of combustion studies. The experiments were conducted in the intermediate-scale fire tunnel, which was designed to simulate environmental conditions in underground mines. The work was divided into three phases. In phase one, the brands were compared for gas production, smoke characteristics, and combustion yields. In phase two, the production constants of transformer fluids and other fuels were compared for the rate of formation of gas and smoke as a function of fire size. In phase three, several diameters of liquid-pool fires were compared in terms of fire size and burning rate. These transformer fluid measurements will be added to the existing coal and wood data that can be used as a basis of comparison for future studies of other mine combustibles. Further research into the combustion product emissions from combustible materials found in underground mines will lead to improved and realistic fire detection and suppression systems,

"The bisbenzylisoquinoline alkaloids are compounds which can be isolated from plants and which exhibit a broad range of pharmacological activities. Perhaps the best known alkaloid of this group is tetrandrine (TT), a compound isolated from the root of a traditional Chinese medicinal herb, Stephania tetrandra. It has been tested for its effects on the cardiovascular system, tumors, and the lungs. For example, tetrandrine has been shown to act as a calcium entry blocker in cardiac sarcolemmal vesicles (King et a/., 1988) and has been used in China as a treatment for angina and hypertension. Its possible effects as a tumor-inhibitory substance have also been investigated (Kupchan and Atland, 1973). In addition, there are several lines of evidence to suggest that it is a possible therapeutic agent for the treatment of silicosis. Yu et a/. (1983) have shown that TT inhibits the development of experimental silicosis in rats when it is administered orally after intratracheal instillation of quartz. Other studies have shown that the alkaloid inhibits the synthesis of collagen by fibroblasts and the formation of silicotic nodules in rats exposed to silica (Huang et al., 1981; Liu et al., 1983). In fact, tetrandrine has been used clinically in China as an antifibrotic agent and has been shown to improve both the appearance of chest radiographs and the measurements of diffusing capacity in patients with silicosis (Li et al., 1981).Since tetrandrine may have a potential use as a therapeutic agent for fibrotic lung diseases, its effects on various pulmonary cells and tissues should be studied. Alveolar type II cells are located in the alveolar regions of the lungs and are important because they are responsible for the synthesis and secretion of pulmonary surfactant (Dobbs et al., 1982; King, 1984), regeneration of the alveolar epithelium after lung injury (Kauffman et al., 1974), and metabolism of foreign compounds (Bend et al., 1985). Furthermore, in silicosis, for which TT has been suggested as a treatment, there are increased amounts of pulmonary surfactant produced (Dethloff et al., 1986a,b) as a result of increased rates of surfactant synthesis by type II cells (Dethloff et a/., 1989; Miller et al., 1988). Because type II cells play a critical role in maintaining normal lung function and because type II cells are affected in silicosis, for which 'TT has been used as a treatment, we studied the effects of tetrandrine on energy metabolism in these cells in vitro. Energy metabolism was chosen because it is a basic cellular function which is neces-sary for other cellular processes to occur. Since type II cells depend on mitochondrial metabolism for energy production (Fisher et al., 1980), we examined the effects of tetrandrine on cellular and mitochondrial ATP production and oxygen consumption. Additional experiments were performed in an effort to determine if intracellular calcium levels may be involved in the TT-induced effects. A preliminary report of those results has appeared previously (Ma et al., 1991)."

The purpose of this study was to assess the adequacy of the cab design and to determine the percentage of musculoskeletal symptoms among operators of mobile equipment used in mining and construction. A questionnaire was designed to assess demographics, work information, job history, and musculoskeletal symptoms in operators of heavy mobile equipment. Information concerning equipment included design of the seat/chair, levers, pedals, bothersome vibration, quality of ingress/egress from the equipment, proper preventative maintenance and repairs, and age of the equipment. The body regions that were evaluated included the neck, middle/upper back, low-back, shoulder/upper-arm, elbow/forearm, wrist/hand, hip, knee, and ankle/foot. Five hundred and eighty six operators completed the questionnaire. The results indicate that these workers are at risk for developing musculoskeletal disorders, and the need to quantify risk factors (i.e., whole-body vibration and static sitting postures).

The main objective of occupational health and safety research is to minimize or eliminate the events that may cause fatal or non-fatal injuries to human workers. A commonly used technique is to devise an “incident prevention plan”, which is more often the product of thorough investigations of past reported incident events. Incident documentation and methodical reporting and systematic and thorough data collection, often help identify the root cause of the incident – in other words, the etiology of the incident. These retrospective analyses of the incidents can efficiently identify the future steps to take to achieve the objective of minimizing occurrence of “events of interest”. This paper introduces a novel technique – “Taxonomic Analysis”, which identifies the root causes of an event (an incident in this case) and can provide future direction for corrective measures to reduce the probability of occurrence of the event. A taxonomic analysis involves systematic and organization of data based on observation, description, and classification. This paper utilizes the rock fall related incident narratives, available with the Mine Safety and Health Administration (MSHA) database (1984-1999), for a taxonomic analysis. The study found that 67.6% of groundfall incidents occurred in supported areas with the remainder (32.4%) in unsupported areas. Some form of rock mass failure appears to cause about 50% of groundfall incidents, although in 39.7% of the overall groundfall incidents or two-thirds of those involving rock mass failure, the exact nature of the rock mass failure could not be determined from the narratives. Human activities factors such as scaling or barring down, drilling or bolting, and setting timbers or cribbing accounted for another 34.5% of the groundfall incidents. Finally, support system failures made up the remaining 15.5%. Results from this taxonomic analysis may suggest focus areas toward which preventive measures and future research activities could concentrate.

[At the majority of mines where flame safety lamps are used either as working lights or as a means of detecting gas, no check it made on this condition of the various parts of the lamp and their assembly other than a visual ink inspection. Although this inspection should be made carefully in view of tae serious consequence that may follow Vie use of a faulty lama, In many instances the inspection is of a perfunctory nature. The Bureau of Mines has therefore designed a small box or gallery for testing lamps for defects and improper assembly before they are taken into the mine. The test simulates the condition when a lamp is extinguished by its introduction into an explosive mixture and the operator attempts to relight the wick while the lamp contains the explosive mixture. This test is more rigorous than merely subjecting a lighted lamp to an explosive methane-air mixture. In the latter test the products of combustion tend to blanket the propagation of flame from within the lamp to the exterior, especially if there are defects at the top of the gauzes. Moreover, when the lighted lamp is subjected to an explosive methane-air mixture the methane builds up slowly within the lamp until the lower inflammable limit is attained, whereupon a feeble explosion occurs within. If the nonlighted lamps is first filled with the methane-air mixture that gives the maximum violence and is than ignited, the dampening effect of products of combustion is obviated and the strain on defects is]

"Introduction.During the past two years the Bureau of Mines has conducted a study of natural-gas heaters*** to determine (1) what types of heaters literate carbon monoxide; (2) why carton monocide is liberated; and (3) how, by change of design or construction, the liberation of this gas can be largely eliminated, In addition to the above study the bureau has investigated cases in the Pittsburgh dis¬trict where fatalities have resulted from carbon monoxide given off by natural-gas heaters.Since these fatalities were first brought to the attention of this bureau through the press, it is likely that some cases were missed and thus not listed. On the other hand, some cases reported as being due to carbon monoxide poisoning, on investigation were found to be due to causes other than carbon monoride.The cases listed herein were shown in all probability to be due to carbon monoxide. Those in which there is any doubt have been omitted; so have those in which there was part asphyxiation, where the victims later recovered."

"The biologic events leading to coal workers' pneumoconiosis (C'WP) are not yet fully understood. There is no good correlation between toxicity studies or animal models with human epidemiologic and pathologic data. Several studies have shown that the severity and prevalence of the CWP in miners correlate strongly with the rank and type of the coal mined. However this has not been found to be the case with laboratory animal models or in vitro studies. In 1980 Russian scientists Artemov and Resnik suggested that the discrepancy may be related to the excessive amount of short-lived free radicals present in freshly generated coal mine dust. Because of their short life, such free radicals would be absent in the ""stale"" coal dust used in most animal experiments. To test this hypothesis we have undertaken a systematic study of the generation, reactivity with oxygen and biological significance of free radicals in coal dust.Electron spin resonance (ESR) was used as a technique for the direct detection of radical production and kinetics of decay. ESR signals from the newly generated radicals decreased with time on exposure to oxygen or when stored in biological buffers at a rapid rate. In vitro biological tests were carried out which suggest that the reaction between the newly generated radicals and oxygen plays a significant role in the sample's cytotoxicity.Materials and MethodsESR measurements were made at X-band (~9.7 GHz) using a computer-controlled Bruker ER200D ESR spectrometer. 11te two coals studied in detail were obtained from the Pennsylvania State University Coal Depository. These were, a bituminous sample, PSOC-1172 (72% carbon) and an anthracite sample, PSOC 867 (95% carbon). Coal particles of several millimeter (mm) dimensions were evacuated to a pressure of ~10-3 mm Hg typically overnight. Without exposure to air, these particles were ground in an agate mortar with an agate pestle for about 30 minutes and then sieved through a 20 \1111 mesh filter before ESR measurements or cytotoxicity studies. As an index of cytotoxicity, hemolysis was measured according to the method of Harrington, with minor modifications. Sheep erythrocytes (2%) were incubated with test samples at 37°C for one hour, and the hemoglobin released was quantitatively estimated photometrically. Additionally, release of alveolar macrophage enzyme lactate dehydrogenase (LOH) was estimated after incubation of the macrophages with the coal particles at different time intervals after the grinding. Enzyme activity was assayed according to the method of Wrobleski and LaDue and expressed as percent of enzyme released in 2 hours per million cells with 1 mg of coal."
