"With a view of elucidating the role of glutathione (GSH) in the biochemical pathways of the chromate-exposure related carcinogenesis, we carried out electron spin resonance (ESR) spectroscopic investigations of the chromate-GSH redox reactions. The ESR measurements, employing spin-traps, provide evidence for the involvement of the glutathione (GS•) radical, as well as an isolable Cr(V)-glutathione intermediate. These results indicater a new mechanism for the reduction of chromate by GSH in cellular environment and help understand the (unexpected increase in Cr(VI)-induced DNA strand breaks at elevated GSH levels.We report here indirect electron spin resonance (EGR evidence for the involvement of the glutathionyl (GS) radical and direct evidence for a Cr(V) intermediate in the reactions of glutathione ¬(GSH) with chromate for the first time.. This work was undertaken to clarify the biochemical pathway of the reduction of chromate by GSH, since this reduction process is thought to be a critical step in the pathogenesis of the chromate-exposure related carcinogenesis. This follows from the literature reports which show that (a) Cr(VI)compounds have been proven to be carcinogens in laboratory animals as well as in human studies2-4, (b) Cr(VI) and not Cr(III) compounds are mutagenic 5-7, (c) Cr(VI) and not Cr(III) compounds can penetrate cell membranes 8.9 (d) the final state of Cr(VI) in cellular reductions is Cr(III)', (e) by itself chromate (or Cr(VI)) cannot be directly mutagenic since Cr(VI) is known not to interact with isolated DNA under physiological conditions 10, suggesting the significance of Cr(VI).reduction processes. One of the major reductants in cellular environments is thought to be the GSH, both outside and inside the ce11s 11-13. Other evidence for the role of GSH in the chromate toxicity is seen from recent studies showing that exposure of hamster cells to non-toxic levels of added selenite increases the levels' of GSH as well as the Cr(VI)-induced DNA strand beaks 14, and thatsuch DNA strand breaks in hepatocytes also change in direct proportion to their GSH content 15,16. These observations wereinterpreted as implying that the reduction of chromate by GSH, to some reactive intermediate is an important step in the chromatecarcinogenicity 15,16. Approach Use the knowledge gained by extensive field tests to design a more rugged and reliable Instrument using lighter and stronger materials. How It Works The original gage consisted of a stainless steel cylinder 1-3/8 Inches In diameter and 10-1/2 inches long. The new gage is made from titanium. This lighter material reduces the weight of the gage by 43 percent, which enhances the gage placement and depth of overcoring in vertical up? holes. For added strength, the titanium case and placement end are now threaded to the gage body instead of using screw fasteners. Three pairs of strain gaged transducers are positioned to measure the diametral deformation of a 1-1/2 diameter borehole at 60-degree intervals around the Inner surface of the borehole. Each transducer consists of a beryllium copper cantilever with a strain gage cemented on each side. These transducers are secured Into the gage body by a tapered shank for minimal creep. The transducers opposite each other are wired together to form a 4-arm external bridge circuit. A pair of transducers is referred to as a component. The performance of Ecuador's mineral industry continued to be affected not only by depressed oil prices in recent years but also by natural disasters. In March 1987, Ecuador canceled foreign debt payments for the rest of the year and appealed for international aid because of a series of earth-quakes that destroyed villages and crippled the nation's oil industry. The earthquake destroyed a key section of the country's main oil pipeline and forced a halt to production and shipment of Ecuador's most important export product. Ecuador produces 240,000 barrels of oil per day and exports 144,000 barrels daily. Oil accounts for 60% of the nation's hard currency earnings. The Government of Ecuador asked the Organization of Petroleum Exporting Countries and other oil-producing countries to assume Ecuador's oil sales commitments. Ecuador agreed to pay the countries back in an equivalent amount of oil when it was able to resume exports.

The Bureau of Mines presents a theoretical solution for the stresses and displacements in an infinite elastic plate containing a circular hole whose boundary is subjected to applied shear loads over four separated arcs. The results for the applied shear loads can be superimposed on the results for applied radial loads, given in a previous publication, to obtain a complete two-dimensional approximation of the stresses and displacements induced in the rock by tunnel boring machines anchored in a circular tunnel by means of four props. Analysis of the various solutions shows that the tangential tensile stresses induced on the boundary of the circular tunnel by applied radial loads can be minimized in the roof and floor of the tunnel by the application of appropriate shear loads to each of the four props. Thus when planning tunnel boring operations, consideration should be given to the in situ stress field in the rock and the induced stresses in the rock resulting from the applied radial and shear loads produced by the props of the boring machine.

Reduce or prevent acid mine drainage from coal refuse piles and surface mines by inhibiting the growth of acid-causing bacteria. Approach A dilute surfactant or deter? gent solution is applied directly to coal refuse piles or overburden using a hydro-seeder or road watering truck. The surfactant treatment can be used either as a preventive measure to avoid a potential acid drainage problem or to reduce water treatment costs by controlling acid drainage at its source. How It Works Acid drainage is prevented or reduced by inhibiting the growth of Thiobacillus ferrooxidans, a type of bacteria which obtains most of the energy it needs to survive by oxidizing ferrous iron in water. The oxidized iron in turn attacks pyrite, which forms an acid and additional ferrous iron for the bacteria to oxidize. T. ferrooxidans is protected by an outer membrane which enables it to survive in its acid environment. Anionic surfactants, or surfactants containing negatively charged ions, can be used to destroy this membrane, thus killing the bacteria and slowing down the oxidation of acid-forming pyrite. Of the anionic surfactants tested to date, sodium lauryl sulfate (SLS) appears to be the most effective as a bactericide. Alpha olefin sulfonate and alkyl benzene sulfonate are acceptable alternatives. These surfactants are major components of common household products, such as laundry detergent, shampoo and toothpaste. They are, therefore, readily available, inexpensive, biodegradable and present no environmental problems at low concentrations.

The periphery of longwall panels in the Pocahontas No. 3 coalbed is characterized by a zone of reduced permeability that inhibits the natural drainage of methane. A residual gas pressure of 105 psig was measured after 1 year in an isolated block of coal measuring 340 by 1,100 feet. The buildup of gas pressure within such large blocks causes methane problems during plowing operations. However, natural drainage holes drilled during development have been found to reduce the gas content of the coal in this coalbed by over 90 percent. The effect of water infusion on dust during plowing operations was investigated using existing methane drainage holes. These holes were cleaned of debris and a pipe was grouted into the hole for water infusion of the panel. Both total and respirable dust levels were reduced by 40 to 79 percent. Water infusion of the panel proved more effective than did the use of a plow-mounted spray sys tem.

8. Refuge Chambers 8.1 Refuge Chambers - Technical Discussion 8.1.1 Introduction As a part of the overall escape system, refuge chambers can provide a last resort when existing escapeways are blocked. These chambers are intended to provide both protection from toxic gases and a life supporting environment for a period of time before external rescue or further escape efforts can be carried out Escapeways may be blocked physically (due to cave-ins, etc), by fire, or by the presence of a toxic or oxygen deficient atmosphere. In the first two cases, the trapped miners would most likely await external rescue efforts, and hence the design of the chamber will have to consider the time durations that such efforts may take, It is also possible, in the second case, for fire fighting and fire protective equipment to be stored in the chamber so that miners may attempt their own escape. In the third case, it should be recognized that a compact, beltmounted 1-hour self-contained breathing apparatus has been developed(9), and that other, less compact equipment is available for up to 4 hours of self-contained breathing supply. If such equipment were stored in the chambers, then miners could escape by travelling from chamber to chamber, taking temporary refuge in each before proceeding to the next. 8.1.2 Background The concept of "barricading" has long been recommended for those emergencies where miners are trapped inby a fire. Barricading, which is the construction of a makeshift shelter in a crosscut or deadend, protects the entrapped miners from toxic gases and enables them to conserve a supply of air. In this manner, they can survive two or three days--hopefully enough time for them to be reached by rescue teams. In the past, many lives have been saved with this technique.

As a part of ongoing research into the sulfidation partitioning of selected complex minerals, the U.S. Bureau of Mines conducted research designed to selectively recover Cu from sulfidized chalcopyrite (CuFeS2). Ferric chloride (FeCI3) solutions leached >90 pct of the Cu and <20 pct (sometimes < 10 pct) of the Fe in the sulfidized concentrates. Cupric chloride (CuCI2) leaches were even more selective, solubilizing essentially 100 pct of the Cu and < 5 pct of the Fe. In contrast, leaching non-sulfidized CuFeS2 with either FeCl3 or CuCl2 showed that somewhat more Fe than Cu dissolved. These studies demonstrate that Cu is substantially more leachable from sulfidized CuFeS2 than from nonsulfidized CuFeS2? The Cu-and Fe-bearing constituents generated as sulfidation products, CuS (covellite) and FeS2 (pyrite), respectively, are differentially dissolved. The solubility of Fe from pyrite in sulfidized CuFeS2 is much lower than the solubility of Fe from complex nonsulfidized sulfide where Fe and Cu are bound in the same crystal lattice. Thus, the Cu dissolves selectively, producing a high-Cu filtrate from which Cu can be recovered by cementation or by electrolysis. Flotation of CuS-FeS2 mixtures indicates partitioned phases can be physically separated if effective liberation can be achieved. Magnetic separation tests yielded less promising results.

Few studies have specifically measured and documented the large-scale loading behavior and durability of ventilation stoppings to mining induced movements, particularly from longwalls. Ventilation stoppings are more commonly the concern of ventilation engineers, and ventilation stopping response to ground movement has only been documented in an incidental manner. This paper presents the investigations of underground measurements that have been conducted to determine the loading response of stoppings constructed from lightweight aggregate concrete masonry units (CMU). These investigations have produced some interesting results that may prove beneficial, not only for developing and assessing alternative stopping construction techniques, but also for designing and selecting standard construction methods for use in varying mining conditions. For instance, the interface friction that results from wedging a CMU stopping during construction plays a substantial role in its ability to resist both lateral and vertical loads. Although they are not intended for ground support, the study showed that block stoppings can resist vertical loads of at least 2,700 to 3,000 kN. Asymmetric loading of a stopping may result in localized failure of blocks within a stopping, which, depending on the severity, can be a precursor of impending stopping failure. In addition to measurement results and implications, the paper will also present details of field measurement methods used to assess stopping response. Stopping stiffness and material strength characteristics will also be presented.

This report analyzes mine fires for all U.S. underground and surface coal mining categories by state and2-year time periods during 1990–1999. Risk rate values are derived, and ignition source, methods of fire detection and suppression, and other variables are examined. The data were derived from Mine Safety and Health Administration (MSHA) mine fire accident publications and verbal communications with mine personnel. The analysis will provide the National Institute for Occupational Safety and Health, MSHA, and the mining industry with a better understanding of the causes and hazards associated with mine fires and will form a basis for future fire research programs.

The Bureau of Mines made a laboratory study of the amenability of anorthosite rock ( from the San Gabriel Mountains , Los Angeles County , Calif . ) to treatment by the lime soda sinter process for the extraction of alumina . The study was done under a cooperative agreement with the California Division of Mines and Geology . This report presents the results of the investigation , and it includes a review of the geology of the deposit ( 7 ) by Dr. Gordon B. Oakeshott . Interest in the recovery of alumina from submarginal aluminum silicate ores is stimulated by the fact that the domestic aluminum industry depends on imports of bauxite for its alumina requirements . Of the 8.0 million long dry tons of high - grade bauxite consumed in 1961 for producing metallurgicalgrade alumina , only 0.9 million long tons , or 11 percent , was mined in this country . During past years many organizations have carried out research programs to determine if alumina could be extracted economically from such domestic ores as clays ( 3 , 4 ) , anorthosite ( 8 ) , and other silicate sources . Successful alumina extraction is the ultimate objective of the Bureau of Mines program.

This report revises an earlier publication by the Bureau of Mines on tungsten analytical procedures and expands the coverage by including appropriate procedures for the critical impurity elements that commonly occur in tungsten ores and concentrates (13).3 The basic methods described are not original, but many of the modifications to improve reliability and sensitivity were developed by the Bureau. Gravimetric and colorimetric methods currently used for the determination of tungsten in ores, mill tailings, and concentrates are described. Colorimetric procedures are preferred for low concentrations of tungsten, whereas either procedure is satisfactory for moderate concentrations. Conversely, a gravimetric method is preferred for the determination of tungsten in concentrates.

Methane in coal constitutes a serious hazard in coal mining. Draining the methane in advance of mining can mitigate this hazard, conserve energy, and offer economic benefits. This Bureau of Mines report evaluates the Pittsburgh coalbed in Greene County, Pa., to determine if advance drainage is feasible. The gas content of virgin reserves in Greene County is estimated at 100 to 200 ft3/ton, a total of over 370 billion ft3 of gas in an area approximately 310 mi2. Because of the similarity to natural gas, drained coalbed gas could be recovered and added to pipeline supplies of natural gas. The value of conserved gas would offset the cost of drainage and extend energy resources.

Cement kiln dust (CKD) is a waste product generated at the rate of 4 to 12 million tons per year in the United States. The dust has considerable resource potential as a neutralizer for coal mine waste effluents and as a lime substitute in the agriculture and construction industries. However, little information has been published on the composition and characteristics of CKD, and a concern arose over the potential environmental effects of the waste when an analysis of a single sample from West Germany revealed lead and zinc concentrations of 5,620 µg/g and 16,200 µg/g, respectively. [In producing crude oils of paraffin base and of mixed base the producer is often confronted with the problem of preventing and removing paraffin that settles out of oil and accumulates in the well. This paraffin consists of heavy waxes with some sand and asphalt, and varies in. consistency from a soft semifluid wax to hard gummy wax. This problem is by no means a new one as troubles have been encountered with it since the earliest days of the industry. However, with the discovery of the panhandle, Texas, field and other large fields in which extensive troubles have been encountered, the problem has assumed added importance. In new fields the most serious trouble with paraffin is in the flowing wells the paraffin accumulates in the casing and tubing of such wells and unless removed will completely plug the well. This condition also develops in wells which are produced by gas lift, and often presents a serious problem when, this method of producing the oil is used. In older fields paraffin is also deposited on the face of the producing sand that on the walls of the hale or within the interstices of the sand near the hole, and this materially affects the amount of oil that can enter the well.]

Background Permanent stoppings or partitions arc used in mines to separate ventilation airways. The ability of stoppings to retain their structural integrity for a minimum length of time under lire conditions and prevent the spread of flame and toxic combustion products to an adjacent airway is critical. The success of miners in reaching safety during a fire depends largely on the presence of uncontaminated airways. Traditionally, permanent stoppings for coal mines are constructed of concrete blocks. Recently, paneled steel stoppings that satisfy many of the requirements demanded of permanent stoppings, such as incombustibility and strength, have been introduced. Their use also introduced concerns such as the ability of a steel stopping to maintain its ventilation control function when exposed to a fire and the potential that the fire could transmit enough heat through the stopping to ignite combustible materials on the unexposed side. To address these concerns, the Bureau of Mines investigated the fire endurance of galvanized steel and concrete block stoppings.

To produce industrial-grade titanium castings by processes that are nonpolluting and lower cost than the currently used rammed-graphite process. Approach Foundry-grade zircon and olivine sands, bonded by waterglass or bentonite, are used to make shell or rammed molds which require relatively low-drying temperatures and do not generate noxious fumes during drying or casting. Procedures Three separate processes have been developed which meet the stated pollution and cost goals. These are: 1. Waterglass-bonded rammed zircon or olivine sand molds are dried at 480°F, zirconia washed, redried, and used in an inductoslag or skull-casting furnace. 2. Waterglass-bonded zircon or olivine sand shell molds are formed at 560°F, cured at 620°F, zirconia washed, dried at 480°F. and used in an inductoslag furnace. 3. Rammed zircon and olivine sand molds are bonded with western bentonite, zirconia washed, dried at 980°F, and used in an inductoslag or skull-casting furnace.

This report describes laboratory-scale experimental treatment of six base-metal ore samples taken by engineers of the Bureau of Mines, Region II, while investigating ore deposits in California and Nevada. The information is of interest in developing the mining districts represented or when similar ores from other sources are being studied. Simple treatment methods were employed as far as possible. More complex treatments also were tried when necessary. The possibilities and limitations of the methods applied to the several ores are shown by the data herein reported.
